Application of Temozolomide (CAS: 85622-93-1)

 

I. Core Clinical Pharmaceutical Application

Temozolomide is an oral alkylating antineoplastic bulk drug and a first-line chemotherapeutic agent for brain tumors. It exerts therapeutic effects via metabolic activation as a prodrug to induce DNA methylation damage:

Glioblastoma (GBM)

  • Newly diagnosed glioblastoma multiforme: Administered concurrently with radiotherapy during chemoradiotherapy phase, followed by single-agent adjuvant chemotherapy, constituting the standard first-line treatment regimen for adult glioblastoma.
  • Recurrent/refractory glioblastoma: Preferred single-agent salvage chemotherapy after failure of prior chemotherapy regimens.

Anaplastic Astrocytoma

Applied as second-line chemotherapy for recurrent anaplastic astrocytoma post-surgery and radiotherapy, as well as systemic chemotherapy for unresectable cases.

Other Central Nervous System Malignancies

Clinically expanded for adjuvant and palliative chemotherapy of malignant gliomas including oligodendroglioma and ependymoma.

Clinical Trial Exploration for Solid Tumors

Under clinical research for combined chemotherapy regimen development against metastatic malignant melanoma, pituitary adenoma, brain metastasis from lung cancer, neuroendocrine tumors and other solid malignancies.
