DODAP (CAS: 127512‑29‑2, full name 1,2-dioleoyl-3-dimethylammonium-propane) is a cationic lipid widely used in biochemistry and drug delivery. Its main functions are:
Gene Delivery Carrier
DODAP can be used to prepare liposomes or nanoparticles for delivering DNA, siRNA, or mRNA into cells.
It forms cationic lipid–nucleic acid complexes (lipoplexes) by binding with negatively charged nucleic acids, protecting them and facilitating cellular uptake.
pH-Sensitive Lipid
DODAP is mostly neutral at physiological pH, reducing toxicity.
In acidic environments (such as endosomes), it becomes positively charged, helping disrupt the endosomal membrane and release nucleic acids into the cytoplasm.
This property makes it highly useful in mRNA vaccines and gene therapy.
Drug Delivery Aid
DODAP can be combined with other lipids (e.g., DSPC, cholesterol, PEG-lipid) to form stable lipid nanoparticles (LNPs) for delivering small molecule drugs or nucleic acid therapeutics.
