1. Key Intermediate for Venetoclax (BCL-2 Inhibitor)

5-Hydroxy-7-azaindole is a critical intermediate in the synthesis of Venetoclax (ABT-199), a highly selective BCL-2 inhibitor developed by AbbVie. Venetoclax is clinically approved for the treatment of chronic lymphocytic leukemia (CLL), acute myeloid leukemia (AML) and other hematological malignancies. As the 7-azaindole core building block, it constructs the key pharmacophore of Venetoclax that binds to BCL-2 protein to induce apoptosis of cancer cells. It is applied in the process development and scale-up production of Venetoclax API.

2. Pharmaceutical Impurity Reference Standard

As an associated impurity (Venetoclax Impurity 12) in the Venetoclax synthetic route, it is applied for impurity profiling, qualitative and quantitative analysis of impurities, analytical method development and validation, and quality control release testing of Venetoclax API and finished drug products.

3. VEGFR-2 Inhibitor Synthesis

1H-Pyrrolo[2,3-b]pyridin-5-ol is also used as a synthetic building block in the development of potent VEGFR-2 (vascular endothelial growth factor receptor 2) inhibitors, which are a class of targeted anticancer agents for anti-angiogenesis therapy. The 7-azaindole scaffold containing a modifiable hydroxyl group enables the construction of diverse kinase inhibitor libraries for anticancer drug R&D.
